FOXO4-DRI is an 18-amino acid D-amino acid retro-inverso (DRI) peptide designed to interfere with the FOXO4–p53 protein-protein interaction that anchors senescent cells in survival. Developed by Peter de Keizer’s laboratory (Utrecht University), it represents a novel class of senolytic tool compound — selectively inducing apoptosis in senescent cells while leaving non-senescent cells unaffected.

Why Researchers Select FOXO4-DRI

Cellular senescence — the state of permanent growth arrest in damaged or aged cells — is maintained partly through pro-survival signals that prevent apoptosis. In senescent cells, FOXO4 (a forkhead transcription factor upregulated in senescence) translocates to the nucleus and interacts with p53, sequestering p53 away from its pro-apoptotic role at mitochondria. FOXO4-DRI competes with endogenous FOXO4 for p53 binding, releasing p53 to activate mitochondrial apoptosis selectively in senescent cells. Published mouse studies demonstrated clearance of p21/p16-positive cells and improvements in fitness, fur density, and renal function markers in aged mice.

Storage Instructions

All our research peptides are manufactured using a lyophilization (freeze-drying) process. This method is designed to maintain product integrity and allows vials to remain stable during shipping for approximately 3–4 months.

Once a vial is reconstituted with bacteriostatic water, it should be stored in the refrigerator to help maintain stability. Under these conditions, reconstituted material is generally considered stable for up to 30 days.

Lyophilization is a dehydration technique in which compounds are frozen and then exposed to low pressure. This causes the water in the vial to sublimate directly from solid to gas, leaving behind a stable, crystalline white structure. This powder can be kept at room temperature until reconstitution.

Upon receipt, products should be stored away from heat and light. For short-term use, refrigeration at approximately 4°C (39°F) is suitable. For long-term storage (several months to years), vials may be placed in a freezer at approximately -80°C (-112°F). Freezing is the preferred method for preserving product stability over extended periods.
