Ipamorelin is a pentapeptide GH secretagogue (Aib-His-D-2-Nal-D-Phe-Lys-NH₂) and selective agonist of the ghrelin receptor (GHSR-1a). Developed as a second-generation GH secretagogue, it was specifically engineered to stimulate GH release through the ghrelin/motilin-related pathway while avoiding the off-target hormonal effects — particularly ACTH and cortisol release — that limited the utility of earlier compounds like GHRP-6.
